Specificity | This antibody is specific for murine VCAM-1. VCAM-1 is an important in cell-cell recognition. Appears to function in leukocyte-endothelial cell adhesion. Interacts with integrin alpha-4/beta-1 (ITGA4/ITGB1) on leukocytes, and mediates both adhesion and signal transduction. The VCAM1/ITGA4/ITGB1 interaction may play a pathophysiologic role both in immune responses and in leukocyte emigration to sites of inflammation. |

Application Notes | The original rat version of this antibody was used to treat experimental autoimmune encephalomyelitis (EAE in female SJL/N mice). It resulted in a later onset of tEAE (Engelhardt et al, 1998; pmid:9854045). To asses α4-integrin–mediated T cell interaction with VCAM-1 on blood-brain barrier–endothelium in experimental autoimmune encephalomyelitis flow cytometry was preformed on T cell blasts labeled with the rat version of this antibody (Vajkoczy et al, 2001; pmid:11518729). Futhermore, PLP- specific T lymphoblasts were used for immunofluorescence with the rat version of this antibody (Vajkoczy et al, 2001; pmid:11518729). And finally, mice were injected with the rat version of this antibody to asses how it can slow down progression of EAE. It showed an ability to slow EAE down (Vajkoczy et al, 2001; pmid:11518729). |

Note on publication | The effects of mAb therapy on the development of EAE in the SJL/N mouse was compared, using a large panel of mAbs directed against alpha4, beta7, the alpha4beta7-heterodimer, and against VCAM-1. Although encephalitogenic T cells express both alpha4-integrins, mAbs directed against the alpha4beta7-heterodimer or against the beta7-subunit did not interfere with the development of EAE. |
