Specificity | This antibody binds mouse FcgammaRIV (CD16a). This molecules acts as a receptor for mouse IgGs and binds with intermediate affinity to both mouse IgG2a and IgG2b. It does not bind mouse IgG1 or IgG3. It recognizes neutralizing virus specific-IgGs displayed on the surface of infected cells and triggers antibody-dependent cellular cytotoxicity (ADCC). |

Application Notes | This antibody binds 293F cells transfected with mFcγRIV with a binding affinity of 4.3nM. This antibody could positively stain mFcγRIV expressed on the surface of macrophages and neutrophils in a flow cytometry (PMID: 26512139). The binding specificity of this antibody towards mouse FcgammaRIV expressed on peripheral blood monocytes from WT or FcgRI, II, III2/2 (TKO) mice and thioglycolate-elicited peritoneal macrophages from WT mice was determined using flow cytometry. This antibody was also capable of binding plate-bound FcgRIV–CD4 fusion protein in an ELISA. The binding specificity of this antibody was further confirmed by immunocytochemistry of fresh frozen spleen sections of naive mouse. Immunofluorescent staining of FcgRIV from thalamus of NBH animals and ME7 animals before and after LPS challenge shows colocalization between FcgR and microglia (PMID: 21572034). This antibody was used in immunohistochemical analysis of mouse retina to study the presence of immune complexes, Fcγ receptors (FcγRs), and markers of macrophage/microglia activation in age-related macular degeneration (PMID: 24334446). |
