The original antibody was generated by immunizing Balb/c mice with synthesized GAβ.
Application Notes
The original antibody was generated and screened in vitro via ELISA and Thioflavin T (ThT) fluorescence measurements for its ability to bind to GAβ and inhibit Aβ polymerization. It was assessed in three polymerization systems (1) polymerization initiated by adding Aβ fibril fragment, (2) Aβ40 polymerization initiated from GAβ40, and (3) Aβ42 polymerization initiated from GAβ42. It showed stronger inhibitory activity, particularly in Aβ40 and Aβ42 polymerization systems, and was found to influence fibril length by promoting fibril shortening or fragmentation rather than just preventing elongation (US7741448B2).
