Specificity
This antibody recognizes a discontinuous conformational epitope located on domain III of the ectoplasmic region of the apical membrane antigen 1 (AMA1) of Plasmodium vivax and also cross reacts with AMA1 from Plasmodium knowlesi, Plasmodium cynomolgi, Plasmodium berghei and Plasmodium falciparum. AMA1 is involved in parasite invasion of erythrocytes.

Immunogen
The original antibody was generated by immunizing BALB/c mice with recombinant ectoplasmic region of AMA1 from Plasmodium vivax.
Application Notes
This antibody can recognize AMA1 of Plasmodium vivax, Plasmodium knowlesi, Plasmodium cynomolgi, Plasmodium berghei and Plasmodium falciparum in an immunofluorescence assay on mature schizonts. The binding region is a discontinuous epitope located on domain III of the ectoplasmic region, the major component being a loop containing a cystine knot. The binding of F8.12.19 to recombinant AMA1 from both P. vivax and P. falciparum was measured by surface plasmon resonance. This antibody shows no function-blocking effect was detected in ex vivo parasite growth inhibition assays. (PMID: 17229439). This antibody a dilution of 1 16,000 was used for the detection of recombinant PvAMA-1 and recombinant PvAMA-DII-III using dot-blot and ELISA assays (PMID: 28855657). This antibody was also used for the identification of PfAMA1 using western blot (PMID: 25236489).
