Specificity
This antibody binds human cytokeratin 18. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. Cytokeratin 18 is widely expressed in human monolayer epithelial tissues, and generally appears in pairs with CK8. CK18 is a serum marker for a variety of tumor diagnosis, as well as a serum marker for liver injury. It is involved in the uptake of thrombin- antithrombin complexes by hepatic cells. When phosphorylated, it plays a role in filament reorganization. It is also involved in the delivery of mutated CFTR to the plasma membrane. Together with KRT8, is involved in interleukin-6 (IL-6)-mediated barrier protection.

Immunogen
The original antibody was generated by immunizing BALB/c mice with recombinant human cytokeratin 18 protein with histidine label in equal volume of Freund's in-complete adjuvant.
Application Notes
The initial binding characterization of this antibody was done using ELISA. The affinity constant of the original IgG1 antibody is reported to be 1.92 × 10^(9) M^(-1). This antibody was used in the immunohistochemical analysis of small intestine and lung carcinoma. The immunohistochemical staining of this antibody is similar to another commercially available antibody called 5D3 (CN113735971).
