Specificity
1B3 antibody recognises digoxin and digitoxin with an excellent binding affinity of Kd = 3 and 0.6 nM, respectively. Digoxin and digitoxin are both cardiac glycosides used widely in cardiology to treat various heart conditions, such as heart failure.

This antibody was raised using transgenic HC2/KCo5 mice with the humanised humoral immune system capable of generating human IgG1 κ Abs.The mice were immunised with digoxin conjugated to keyhole limpet hemocyanin (KLH).
Application Notes
Digoxin is widely used in various heart conditions. However, the drug has a very narrow therapeutic index and can cause toxic side effects. The antibody might neutralise the activity of digoxin and could be used as an antidote in case of overdose of digitalis (Paula et al., 2005). It is additionally superior to the widely utilised Fab preparations of sheep polyclonal anti-digoxin Abs as it is a humanised antibody which will not activate the immune system in patients (Ball et al., 1999). Anti-digoxin monoclonal antibodies can also be used to measure serum levels of the drug as a guide to therapy, and are useful in the investigation of the kinetics of interaction between antibody and antigen.
