Specificity
This antibody binds human DLL3 in the N-terminal domain and also reacts with mouse, rat and cynomolgus monkey DLL3. Delta-like protein 3 (DLL3) is a transmembrane protein that belongs to the Delta/Serrate/Lag-2 (DSL) family of Notch ligands. Mature human DLL3 consists of a 466 amino acid (aa) extracellular domain (ECD) with one DSL domain and six EGF-like repeats, a 21 aa transmembrane segment, and a 105 aa cytoplasmic domain. It is an inhibitory Notch ligand that is highly expressed in small cell lung cancer (SCLC) and other neuroendocrine tumors but minimally expressed in normal tissues. It is therefore being explored as a potential therapeutic target in SCLC.

Immunogen
The original antibody was generated by immunizing three strains of mice (Balb/c, CD-1 and FVB) with recombinant human DLL3-Fc or with human DLL3-His fusion constructs.
Application Notes
The binding characterization of this antibody to human DLL3 was done using ELISA. Binding affinity of mouse IgG antibody for human DLL3 is Kd= 0.2 nM. This antibody can bind human, cynomolgus monkey, rat and mouse DLL3 expressed on surface of 293 cells via flow cytometry. This antibody can mediate killing of tumor cells expressing DLL3 and facilitate the delivery of cytotoxic agents to live cells, in an vitro cell killing assay. This antibody was capable of killing OV26 cells, an ovarian NET NTX tumor, with a sub-picomolar EC50 (e.g. 0.14 pM). Humanized version of this antibody is also available on request. Human IgG1 version was capable of killing LU37 cells in an in vitro killing assay. This antibody was used for the generation of antibody-drug conjugates for suppressing tumor growth (US9089615). It was also used in combination with OKT3 for generation of a bispecific antibody targeting DLL3 and CD3. It was also used for the generation of DLL3-targeted CAR-T cells (PMID: 32554616).
