Specificity
2D73 reacts with residues 309-319 (DKEMAETQHGT) within domain III of the DENV envelope protein, shown crystallographically (Li et al, unpublished). Although raised against DENV-3 EDIII, 2D72 cross- reacts with EDIII from all four DENV serotypes. 2D73 does not react with other DENV-related flaviviruses, such as WNV, YFV and JEV.

Immunogen
BALB/c mice were immunised with recombinantly-produced envelope protein domain III (EDIII) from DENV-3. Splenocytes were obtained from immunised mice and fused with the NS-1 myeloma cell line to generate hybridomas.
Application Notes
2D73 was able to strongly neutralise DENV infection in a serotype-independent manner, shown using both ELISAs and ELISPOT-based assays (Li et al, 2013). 2D73 has also been used, as a Fab fragment, to crystallise the DENV-4 EDIII protein (Li et al, unpublished).
