Specificity
This antibody is specific for gastrin 17. Gastrin is a peptide hormone that stimulates the stomach mucosa to produce and secrete hydrochloric acid and the pancreas to secrete its digestive enzymes. It also stimulates smooth muscle contraction and increases blood circulation and water secretion in the stomach and intestine. It has also been implicated in the development or maintenance of various types of cancer, such as pancreatic and stomach carcinoma.

The original antibody was generated by panning three against human antibody phage display libraries using a short 10 amino acid peptide from the N terminal portion of Gastrin 17.
Application Notes
To determine the efficacy of gastrin neutralization by these antibodies in vitro, a flow cytometric measurement of intracellular calcium flux was performed using the human IgG1 version of this antibody on a CCK2R overexpressing cell line (AGS-TR). This antibody showed a 70%-80% neutralization in Ca flux in cells stimulated with gastrin (US8278421) (Hsu et al, 2008; pmid:18589401). Further the binding affinity of the human version of this antibody was determined using surface plasmon resonance and affinity constants were calculated from the kinetic rate constants for antibody binding to biotinylated peptide immobilized on a Streptavidin sensor chip (US8278421) (Hsu et al, 2008; pmid:18589401). The human version of this antibody has shown the ability to increase the stomach pH of mice to pH 4 in combination with human gastrin and famotidine. The pH with only human gastrin and famotidine was pH 2.5 (US8278421) (Hsu et al, 2008; pmid:18589401). Screening of gastrin positive antibody clones was done using ELISA (Hsu et al, 2008; pmid:18589401). Inhibition of gastrin activity has potential for therapeutic use as a suppressor of acid secretion as well as an inhibitor of gastrin-responsive tumors (Hsu et al, 2008; pmid:18589401).
