Specificity
YC15 was shown to bind ectopically-expressed gH on transfected cell lines, as well as neutralise infection by KSHV virions (unpublished observations). Kaposi's sarcoma is a rare type of cancer, characterised by neoplasmic growth, often associated with AIDS.

Antibodies were produced from a murine hybridoma.
Application Notes
The YC15 variable domain sequences were cloned and redesigned into the scFv format (Cai & Berger, 2011), and further fused to PE38, a construct of Pseudomonas aeruginosa exotoxin A which is lacking the N-terminal cell adhesion domain. In this format, YC15 showed specific dose-dependent binding to surface-expressed gH using flow cytometry. As an immunotoxin with the fused P. aeruginosa PE38 construct, YC15 facilitated potent cytotoxic activity based on recognition of surface gH epitopes.

Note on publication
Describes the reformatting of YC15 variable domains (obtained in an unpublished study) as a YC15-scFv-PE38 immunotoxin. The gH-binding properties of YC15 were shown to be retained.
