Specificity
This antibody binds human glypican 3 (GPC3).Glypican-3 is an oncofetal antigen that belongs to the glypican family of glycosyl-phosphatidylinositol-anchored heparin sulfate proteoglycans. It negatively regulates the hedgehog signaling pathway and positively regulates the canonical Wnt signaling pathway. Multiple studies have revealed that GPC-3 is overexpressed in hepatocellular carcinoma (HCC).

Immunogen
The parental mouse antibody GPC3#4 was generated by immunizing BALB/c mice with 50μg of purified human GPC-3 antigen emulsified in Freund's complete adjuvant. This original antibody was generated be grafting CDRs of the parental mouse antibody onto human framework region.
Application Notes
The binding specificity of this antibody for human glypican 3 was determined using ELISA. The original IgG1 version of this antibody binds human GPC3 with an affinity of Kd = 2.84 pM. This antibody can bind GPC3 expressed on the surface of PLC/PRF/5 and Huh7 cells in an in vitro binding assay. This antibody also positively shows ADCC activity on PLC/PRF/5 cells expressing glypican 3 (CN109988240).
