The original antibody was generated by immunizing BALB/c mice with of purified placental αvβ3.
Application Notes
αv-Integrin is an integral membrane protein involved in cell adhesion. Antibody screening by ELISA revealed the antibody reacted specifically with αvβ3. In ELISA on fixed cells (‘CELISA’) the antibody showed a reaction pattern consistent with its recognition of the αv integrin chain (cells expressing αvβ3 and αvβ5 (M21)). The ability of the antibody to recognize native αv integrins was tested by FACS analysis, where the αv-expressing cells (M21) reacted strongly with the antibody. Immunoprecipitation from surface labelled cells showed the antibody detected αv integrin in M21 cells. Furthermore, αv-integrins can function as receptors for vitronectin. However, the antibody had the ability to disrupt stable interaction between vitronectin and αvβ3, and blocked the growth of M21 tumors in nude mice.
