The original antibody was generated through the construction of a phage-displayed single- chain fragment variable (scFv) library from mice splenocytes immunized with specific mimotope peptides. The library was subjected to three rounds of biopanning on G12V and G13D peptide-coated plates.
Application Notes
The original format of this antibody (scFv) was screened against WT and mutated G12 and G13 KRAS and shown to specifically bind mKRAS G12V in an ELISA. The scFv format of this antibody was fused to mHALT-1 toxin and exhibited cytotoxic effects on SW-480 colorectal cancer cells with IC50 of 25.39 μg/mL, with minimal cytotoxicity effect on NHDF cells. The scFv format of this antibody was fused to enhanced GFP and used for IF (Teo et al., 2021; PMID: 33959410).
