Specificity
This antibody is specific for LAG-3. LAG-3 is an immune checkpoint receptor found primarily on the surface of T cells, including regulatory T cells and exhausted T cells. It plays a critical role in downregulating immune responses. LAG-3 is structurally similar to the well-known CTLA-4 and PD-1 checkpoint receptors and binds to MHC Class II molecules, transmitting inhibitory signals to the T cells. This dampens the immune response, helping to maintain immune homeostasis and prevent overactivation, which could lead to tissue damage or autoimmune diseases.

The original antibody was selected from a camel-derived nanobody phage display library by screening against biotinylated LAG-3.
Application Notes
The specificity of the original antibody (camel VHH) was verified via ELISA and WB. In an in vitro T cell and Daudi cells co-culturing experiment, the addition of 200 μg/ml of a human IgG Fc fused version of this antibody led to the best killing effect on tumor cells. Furthermore, in a follow-up experiment, the antibody was shown to promote the killing of tumor cells by T/CD19 CAR-T cells without distinguishing tumor cell types when tested on K562 and Daudi cells (CN114437219A).
