Immunogen
LHRH diacetate tetrahydrate was conjugated to potato acid phosphatase and an emulsion composed of the conjugate and Freunds complete adjuvant was injected into the hind limb of female CAF1 mice. Immune spleen cells were fused with P3-X63-Ag8.653/3 myeloma cells by a modification of the original cell fusion method using polyethylene glycol. A hybridoma screening assays were used to select for high affinity monoclonal antibodies.
Application Notes
SMI-41 has been used in immunocytochemistry experiments to detect the expression of mature LHRH in the brain detect cells containing mature LHRH peptide (Avigdor 2004). SMI-41 also has been expressed in mice via injection of a replication-incompetent, recombinant adeno-associated virus designed to express the antibody. This has been used to cofer long-term infertility in male and female mice (Li 2015).
