Immunogen
The original antibody was generated by immunization of Llama glama with mCherry fluorescent protein. A VHH cDNA library was generated by high-throughput DNA sequencing of a marrow lymphocyte from an immunized llama. Later on this was combined with mass spectrometric (MS) identification of high affinity VHH regions derived from serum of the same animal.
Application Notes
This antibody can be used to detect mCherry fluorescent protein and DsRed in an ELISA. This antibody binds immobilized mCherry with a binding affinity of Kd= 0.18 nM. The Alexa Fluor® 488 conjugated version of this antibody was used to immunofluorescently stain S. cerevisiae expressing mCherry-tagged histone H2B (PMID: 25362362). This antibody was used for the visualization and manipulation of red fluorescent labelled proteins in live cells by using nanobodies targeting fluorescent protein tags (PMID: 31122229).
