Specificity
This antibody binds methotrexate, formerly known as amethopterin. It is a chemotherapy agent and immune-system suppressant. It is often used in treatment of cancer, autoimmune diseases, ectopic pregnancy and for medical abortions.

The original antibody was generated by immunizing a female llama with methotrexate–BIP conjugate.
Application Notes
The binding reactivity of this single chain antibody for methotrexate-BSA was done using direct and competitive ELISA, with no cross reactivity with only BSA. The binding affinity of the original VHH version of this antibody as determined by BIAcore analysis was reported to be Kd=29 nM (PMID: 17011035).
