Specificity
This antibody binds human PD-L2, expressed on dendritic cells, IFN-gamma-treated monocytes, placental endothelium and medullary thymic epithelial cells. This protein is an immune checkpoint receptor ligand which plays a role in negative regulation of adaptive immune response. It is involved in the costimulatory signal, essential for T-cell proliferation and IFNG production in a PDCD1-independent manner.

Immunogen
The original antibody was generated by immunizing 2 alpacas with human PDL2 antigen in Freund's adjuvant. The peripheral blood of the alpaca was taken to generate a yeast display nanobody library and the particular clone was isolated by screening the library against human PDL2. Later on the single domain antibody was humanized using framework grafting method.
Application Notes
This antibody binds human PD-L2 with a binding affinity of Kd= 1.68 nM. The binding ability of this antibody towards human PD-L2 expressed on the surface of CHO cells was determined using flow cytometry. This antibody is capable of blocking the interaction between PDL2 and PD-1. This antibody in combination with an anti-PDL1 antibody 14-02-3 was used in the generation of a bispecific antibody targeting human PDL1 and PDL2 simultaneously. The resulting bispecific antibody was capable of blocking binding of PD-L2/PD-L1 to PD-1 and also block PDL1/PDL2/PD1/luc signaling pathway (WO2021197358).
