Immunogen
The original antibody was generated by immunizing a female dromedary (Camelus dromedarius) with purified prostate-specific antigen mized mixed in Gerbu adjuvant. A VHH library was constructed and the antibody was isolated by panning against PSA.
Application Notes
The specificity of the original format of the antibody was confirmed by ELISA analysis. Enzymatic activity of PSA in the presence or absence of the antibody was tested. The binding affinity of the antibody with three isoforms of PSA (PSAfree, PSA non-ACT-binding, and PSA-ACT complex) were measured by surface plasmon resonance (Kd= 0.16 nM, 0.18 nM and 0.18 nM respectively) (Saerens et al. 2004; PMID: 15459193). The antibody was employed in a surface plasmon resonance biosensor for PSA detection and proved to be a promising method as it enabled the detection of PSA levels as low as 1 ng/mL (Huang et al., 2005; PMID: 16076438). Further, the antibody was employed in a SPR immunosensor for the detection of PSA. The results showed that the biosensor was able to detect PSA at concentrations below 1 ng/ml in 15 min (Saerens et al., 2005; PMID: 16316161).
