Immunogen | The original antibody was generated by immunizing BLAB/c mice with cells of metastatic melanoma SK MEL 23 cell culture and boosted again four weeks later. |
Application Notes | The disialoganglioside GD2 is expressed on the surface of cells of most human melanomas but not on the surface of melanocytes. The specificty of this antibody towards ganglioside antigens GD2 and GD3 was demonstrated using solid-phase radioimmunoassay and enzyme-linked immunosorbent assay as well as by immunostaining on thin-layer chromatography plates using structurally characterized gangliosides. Binding of antibody to live cells was done by flow cytometry and by immunoperoxidase staining on frozen tissue sections. The binding patterns correlated with antitumor activity in antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity assays with human effector cells and complement in an release assay using cell lines derived from the same individual. (PMID: 3815333). This antibody was also used in monitering the levels of GD2 and GD3 in the sera of patients suffering from melanoma and healthy adults (PMID: 2778837) |
