Specificity | This antibody recognizes the Leu7, which is an oligosaccharide antigenic determinant expressed on a peripheral blood subpopulation of ~15% of lymphocytes that comprises almost all the natural killer (NK) and killer (K) cells but not other blood cells like monocytes, granulocytes, erythrocytes, and platelets (PMID:P 6790607). This antibody is also capable of recognizing Myelin-associated glycoprotein (MAG) which is a quantitatively minor component in both peripheral and central myelin sheaths that is thought to have a role in cell–cell interactions within the nervous system (PMID: 6196641). |

Immunogen | The original antibody was generated by immunizing mice with a membrane antigen from the cultured T cell line, HSB-2. |
Application Notes | Indirect immunofluorescence was used to determine the reactivity of this antibody with certain human cultured T cell lines (HSB-2 and MOLT-4 but not MOLT-3) but not with other lines of B cell or phagocytic cell origin. This antibody reacted with 15.1 +/- 7.1% of normal blood lymphocytes but was unreactive with monocytes, granulocytes, erythrocytes, and platelets. This antibody was also used to separate HNK-1+ cells using flow cytometry (PMID: 6790607). The original mouse antibody was used in the immunoperioxidase staining of sections of spleen, tonsils, and lymph nodes where it has been specifically located within the germinal centers, while in the thymus few scattered cells were identified as HNK-1 antigen positive (PMID: 6195296). This antibody could also recognize an antigenic determinant (MAG) of human central and peripheral nervous system white matter by immunoperoxidase staining of tissue sections (PMID: 6196641). |
