Application Notes | This antibody was used as an inhibitor to study platelet adhesion to collagen VI and ECM. It was found to inhibit the binding of von Willebrand factor (VWF) to collagen I and III (Hoylaerts et al., 1997; PMID: 9164855). This antibody was found to have a comparable affinity for baboon vWF as for human vWF and inhibited the binding of baboon vWF to collagen with an inhibitory concentration of 50% (IC50) of 3.5 μg/ml. Furthermore, it prevented the binding of VWF to collagen and removed bound VWF from a collagen surface, albeit with a lower efficacy. Moreover, the antithrombotic efficacy of the antibody was evaluated in a modified Folts model in baboons; this antibody was used to inhibit the interaction between collagen and vWF to prevent arterial thrombosis in baboons (Wu et al., 2002; PMID: 11986216). The crystal structure of the antibody's Fab fragment, in complex with the vWF A3 domain, was determined (Staelens et al., 2006; PMID: 16314412). |
