Name | Rabbit anti-nNOS antibody(N-Terminal) |
中文名字 | 兔抗nNOS抗体;兔抗神经元一氧化氮合酶 |
Description | The N-terminal neuronal nitric oxide synthase antiserum was quality control tested using standard immunohistochemical methods. The antiserum demonstrates strongly positive labeling of rat hypothalamus, striatum, cortex and spinal cord using indirect immunofluorescent and biotin/avidin-HRP techniques. Recommended primary dilutions are 1/1,000 – 1/2,000 in PBS/0.3% Triton X-100 – biotin/avidin-HRP. |

APPLICATION | |
Quality Control | The N-terminal neuronal Nitric Oxide Synthase antiserum was quality control tested using standard immunohistochemical methods. The antiserum demonstrates significant labeling of rat hypothalamus, striatum, cortex and spinal cord using indirect immunofluorescent and biotin/avidin-HRP techniques. By western blot analysis of brain homogenates the antibody specifically labels a band of approximately 155 kD. Immunolabeling is completely abolished by preadsorption with synthetic human nNOS (134-148) at 5 mg per mL of diluted antibody. No cross reactivity with other forms of NOS was observed. |
Tissue | Rat hypothalamus, striatum, cortex and spinal cord |
Perfusion Fixation | • Fixative - 4% paraformaldehyde in 0.1M phosphate buffer, pH 7.4; 500 mL over 20 min. |
Absorption Control | |
Sections | 10 µm cryostat or 50 µm vibratome |
Tissue Incubation | 18–24 hours at 2°–8°C |
Detection System | Use Bn/Av-HRP reagents as instructed by the manufacturer. |
Suggested Dilution | 1/10,000–1/15,000 in PBS/0.3% Triton X-100 - Bn/Av-HRP immunohistochemistry |
NOTES | |
Special Instructions | It is recommended that the researcher perform a primary antibody dilution series using our dilution recommendations as a guideline. Note that a change in the fixation or buffering system from our protocol may change the configuration of the protein which could alter the reactivity with the tissue tested. Please read the instruction booklet carefully before beginning the procedure. |
Concentration | Not applicable. Antibody concentration is only relevant for purified antibodies. |
Storage | After reconstitution, use immediately or refrigerate at 2º–8ºC up to 2 days. For long-term storage, aliquot antibody and freeze at -15°C or lower. Avoid repeated freeze/thaw cycles |
