One is used as coating antibody. The other is conjugated to HRP and used for detection. Standards and diluted samples (100 μl) are incubated in the coated microtiter wells for 45 minutes. After washing the wells, HRP-conjugate (100 μl) is added and incubated for 45 minutes. If HSP70 molecules are
present, they are sandwiched between the capture and detection antibodies. The wells are then washed to remove unbound HRP-conjugate. TMB is added and incubated for 20 minutes. If HSP70 is present, a blue color develops. Color development is stopped after 20-minutes by addition of Stop Solution, changing the color to yellow. Absorbance is measured at 450 nm. The concentration of HSP70 is proportional to absorbance and is derived from a standard curve.
