Specificity
This antibody is specific for the V600E mutation of the BRAF protein kinase. BRAF V600E mutation can promote the methylation of the thyrotropin receptor (TSHR) gene promoter, resulting in silencing of TSHR or a significant decrease in expression, and feedback increase of thyrotropin (TSH), which promotes the growth of tumor cells. BRAF V600E gene mutation and telomerase reverse transcriptase (TERT) promoter mutation also play an important role in the occurrence of papillary thyroid carcinoma (PTC).

Immunogen
The original antibody was generated by immunizing mice with double repeat recombinant BRAF V600E protein 'GDFGLATEKSRW' and BRAF V600 protein 'GDFGLATVKSRW' in equal amount of complete Freund’s adjuvant.
Application Notes
This antibody can be used for the detection of the BRAF V600E mutation. This antibody is useful in the diagnosis and targeted therapy for papillary thyroid carcinoma. The binding characterization of this antibody for BRAF V660E mutation was done using ELISA. The IgG1 version of the antibody binds the BRAF V600E mutation with a dissociation constant of 0.04 nM and IC50 of the IgG1 antibody against A375 cells was 42nM. This antibody was used in the generation of immunomagnetic beads for the detection of BRAF V600E mutation (CN111285935).
