Immunogen
The original antibody was generated by immunizing BALB/c mice with second extracellular loop of the C3aR (amino acid residues 173–332) fused to glutathione-S-transferase (GST). A combinatorial phage library was generated expressing scFv antibodies and panned against second extracellular loop of the C3aR, fused to the maltose binding protein as antigen.
Application Notes
The original scFv antibody binds to the 2e loop of the human C3a receptor in an ELISA. Human C3a receptor is a G protein-coupled receptor stimulating chemotaxis, superoxide anion generation and granule enzyme release. This antibody was able to bind RBL-2H3 cells stably expressing the C3aR and to the human mast cell line HMC-1 in a flow cytometric assay. This antibody is not capable of blocking the interaction between C3a and C3aR (PMID: 11373092).
