Specificity
This antibody is specific for human claudin-18.2 with no cross reactivity with claudin 18.1. Alterations in claudin expression can impair tight junction function, influence signaling pathways, and act as a tumor-promoting event in some epithelial cancers. Claudin 18.2 is overexpressed in various tumor type and is found to be a potentially important target in advanced gastric cancer.

The original antibody was generated by grafting CDRs of the parental mouse antibody onto human framework regions.
Application Notes
The binding specificity of this antibody to human Claudin 18.2 expressed on the surface of HEK293 cells and Kato III cells was done using flow cytometry. The in-vivo anti-tumor activity of this antibody was evaluated in a syngeneic Mc38-huCLDN18.2 xenograft model in mice, which is a murine claudin18.2-/-mouse colon cancer cell line that has been engineered to expresses human claudin 18.2 (WO2021047599).
