Specificity
This antibody is specific for the intermediate chain of dynein. Dynein acts as one of several non-catalytic accessory components of the cytoplasmic dynein 1 complex that are thought to be involved in linking dynein to cargos and to adapter proteins that regulate dynein function. Cytoplasmic dynein 1 acts as a motor for the intracellular retrograde motility of vesicles and organelles along microtubules. The intermediate chains mediate the binding of dynein to dynactin via its 150 kDa component (p150-glued) DCTN1. May play a role in mediating the interaction of cytoplasmic dynein with membranous organelles and kinetochores.

This antibody was raised by immunizing mice with bovine brain cytoplasmic dynein.
Application Notes
The murine version of this antibody bound to protein A-Sepharose beads is able to immunoprecipitate dynein. A western blot on sucrose density purified dynein using the mouse version of this antibody has proven the specificity of this antibody (Dillman and Pfister, 1994; pmid:7528220). A western blot on the intermediate chain of dynein using the mouse version of this antibody determined the interaction between the dynein intermediate chain (IC) and the dynactin p150 (Siglin et al, 2013; pmid: 23577064). The ability of this antibody to inhibit the CC1/NudE-Dynein interaction was determined. To do this the mouse version of this antibody was incubated with rat brain purified cytoplasmic dynein and then mixed with bacterially expressed GST-CC1 or GST-NudE. The results were analyzed using coomassie- stained gels (McKenney et al, 2011; pmid: 21911489). The influence of this antibody on microtubule polymerization was determined in vitro. Cells were grown on cover slips and then incubated with the murine version of this antibody (Zhapparova et al, 2007; pmid: 18205606). An assay was preformed on PtK2 cells which were microinjected with the function-blocking mouse version of this antibody. This determined whether dynein is the cortical tether for microtubules at adherens junctions (Ligon et al, 2007; pmid: 17550375). A western blot using the murine version of this antibody on the intermediate chain of dynein determined the role of cytoplasmic dynein in mitosis and the intracellular transport of organelles, proteins, and mRNAs. Further the mouse version of this antibody bound to protein G dynabeads is able to immunoprecipitate a dynein GPF complex (Zhang et al, 2013; pmid: 23475693). A western blot on Xenopus oocyte lysates using the mouse version of this antibody determined the influence of dynein in the directional transport in a RNA localization pathway. Further the mouse version of this antibody is able to immunoprecipitate proteins from lysated Xenopus oocytes. Finally the mouse version of this antibody was used to stain dynein in Xenopus oocytes during a immunofluorescence experiment (Gagnon et al, 2013; pmid: 23637574).
