Specificity
This antibody is specific for human factor VIII, a glycoprotein involved in blood coagulation. Factor VIII acts as a cofactor for factor IXa which converts factor X to the activated form Xa in the presence of calcium and phospholipids. Defects in the F8 gene, which encodes Factor VIII, result in the coagulation disorder haemophilia A.

Immunogen
This antibody was raised by immunising BALB/C mice with highly purified procoagulant factor VIII (VIII: C). Spleen cells were then isolated from immunised mice, and fused with P3 NSI mouse myeloma cells to generate hybridomas.
Application Notes
This antibody is a potent coagulation inhibitor (Rotblat, 1983).This antibody has been employed to produce factor VIII-deficient plasma by immunodepletion (Takase et al, 1987); purified monoclonal antibodies were bound to Sepharose columns, which were used to reduce the factor VII content of plasma through column chromatography. This antibody does not bind antigens if CRM negative haemophilic plasma, which lacks factor VIII, is used in column chromatography (Rotblat et al, 1983). This antibody has also been used in Western blotting to identify factor VIII (Rotblat et al, 1985)
