Specificity
The antibody binds an epitope in gH D-IV, proximal to the viral membrane and distant from known receptor binding sites. In particular, the antibody binds to the back side of gH D-III/D-IV close to gH D- III residues 494–503 (loop between 3α-8 and 3α-9), 456–468 (3α-6 helix), and 406–415 (loop between 3α-3 and 3α-4), and gH D-IV loop residues 645–656 (loop between 4α-1 and 4β-9), 623–626 (loop between 4β-7 and 4β-8), and 568–577 (loop between 4β-3 and 4β-4).

Application Notes
The antibody blocks EBV-mediated membrane fusion with both B cells and epithelial cells. Flow cytometry cell counts showed that Fab fragment inhibits virus entry. Neutralization experiments with epithelial cells using an EBV-GFP reporter virus and purified Fab. Fluorescence images obtained at 24 h postinfection and flow cytometry cell counts showed that the Fab inhibits virus entry. The Fab exhibited ∼50% inhibition of EBV infection at 150 nM, where no inhibition of cell–cell membrane fusion was observed. (PMID: 28939750) The cell-surface expression of gHgL was evaluated using the antibody by CELISA. (PMID: 27929061)
