Specificity
This antibody binds the gintexin A type PLA2 protein found in the venom of Gloydius intermedius. Gintexin A is Asn-49 type PLA2 isomer that has no enzymatic activity and neurotoxicity. Gentexin is a heterodimer comprising gintexin A and gentexin B. It is estimated that, after the venom enter the blood system GintexinA acts as a "director" to guide the neurotoxin heterodimer to bind to the membrane receptor at the phrenic nerve NMJ synapse, and then Gintexin B exerts neurotoxicity (blocks the diaphragm NMJ synaptic transmission and stops breathing).

Immunogen
The original antibody was prepared by immunizing BALB/c mice with attenuated Gintexin antigen purified from the crude venom of Gloydius intermedius.
Application Notes
This antibody was capable of recognizing attenuated gintexin A of Gloydius intermedius and a fusion protein containing gintexin A in a western blot assay. The binding affinity of this antibody for gintexin A was determined using ELISA. 100 μg this antibody can neutralize at least 2×LD50 of Agkistrodon venom. In vivo, the neutralizing activity of this antibody is reported to be about 21.1 LD50/mg. The sensitivity of HRP-N20 antibody for immunospot detection of Agkistrodon intermedius venom is 12.5 ng/mL.
