This antibody recognizes the active (two-chain) tcHGF but not the inactive (single-chain) scHGF. It does not cross-react with mouse HGF.

The original antibody was generated by immunizing MRL/lpr mice with tcK2SP(Xa).
Application Notes
The specificity of the original format of the antibody for HGF was confirmed by ELISA analysis. The antibody weakly inhibits the HGF activity only at low concentration range. The binding affinity of the antibody toward the HGF proteins was tested, and obtained KD values of 2.19 nM and 1.10 nM for tcK2SP(Xa) and tcK4SP(Xa), respectively. The antibody could specifically immunoprecipitate tcHGF (Umitsu et al., 2016; PMID: 27608665). The crystal structure of the Fab fragment of the antibody in complex with the active form of HGF was determined. The antibody selectively recognized tcHGF in western blot analysis. The antibody was used for immunohystochemistry staining of HGF in fore and hind-stomach and in the corpus epithelium of the adult stomach (Jangphattananont et al., 2019; PMID: 31212972).
