Specificity
This antibody binds mouse Interleukin-1 beta, which is a potent pro-inflammatory cytokine. Initially discovered as the major endogenous pyrogen, induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production. It promotes Th17 differentiation of T-cells and synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells. It also plays a role in angiogenesis by inducing VEGF production synergistically with TNF and IL6.

Immunogen
The original antibody was generated by immunizing IL-1α/β double KO mice with mouse IL-1 beta.
Application Notes
The original mouse IgG antibody binds mouse IL-1B with a binding affinity of Kd= 0.23 nM. The binding characterization of this antibody was done using ELISA. In vitro antibody potency was determined by its ability to inhibit IL-8 production by MRC-5 cells. This antibody was able to inhibit IL-8 production by MRC-5 cells with an IC50 of 3.70E−10M. This antibody was used in combination with 9H10 (anti- mouse IL-1A) antibody for the generation of anti-mouse IL-1α/β DVD-Ig molecule (US7612181).
