Immunogen
The original mouse version of this antibody was raised by immunizing mice with purified human alpha5beta1. Integrin alpha-5/beta-1 (ITGA5:ITGB1) is a receptor for fibronectin and fibrinogen. It recognizes the sequence R-G-D in its ligands. ITGA5:ITGB1 binds to PLA2G2A via a site (site 2) which is distinct from the classical ligand-binding site (site 1) and this induces integrin conformational changes and enhanced ligand binding to site 1. ITGA5:ITGB1 acts as a receptor for fibrillin-1 (FBN1) and mediates R-G-D- dependent cell adhesion to FBN1.
Application Notes
The murine version of this antibody was used in a HUVEC direct binding assay. In this assay the ability of the antibody to bind to HUVACs was tested (US2012195908). Furthermore, the murine version of this antibody was also utilized in flow ctyometry to stain RAJI cells and HUVEC cells. It only bound HUVECs and not RAJI cells (US2012195908). Additionally, it was determined that the murine version of this antibody could inhibit cell adhesion to fibronectin (US2012195908). An HUVEC Caspase-3/7 Activity Colometric Assay was preformed using the murine version of this antibod to determine the apoptosis resulting from the administration of the antibody (US2012195908). HUVEC tube formation assays were preformed using the murine version of this antibody (US2012195908). The humanized version of this antibody was shown to bind to human recombinant integrin α5β1 in ELISA (US2012148572). Furthermore, the binding affinity of the humanized version of this antibody was confirmed using surface plasmon resonance (US2012148572). Finally, a skin wound healing assay was preformed. The humanized version of this antibody showed inhibition of wound healing in white new zealand rabbits (US2012148572).
