The original antibody was isolated from an scFv phage display library generated from BMV (bone marrow), CS (spleen) and DP47 (frame work based CS) and by panning against a fusion protein containing Notch3 extracellular fragment (amino acid 40-467) and human IgG1-Fc.
Application Notes
The original human IgG1 antibody hN3S2-160 binds notch 3 with an EC50 =0.06 nM. The binding affinity of this antibody for notch 3 was determined using ELISA. The binding of hN3S2-160 to notch 3 on 293 Flp-TRex cells was done using flow cytometry. This antibody is also capable of blocking notch 3 and DLL4 ligand interaction (WO2010141249).
