Specificity
This antibody is specific for NTF2. Its reactivity in humans, mice, rats, monkeys, hamsters, chickens and xenopus have been proven. The epitope of this antibody lies in the extreme C terminus of the NTF2 protein. NTF2 mediates the import of GDP-bound RAN from the cytoplasm into the nucleus which is essential for the function of RAN in cargo receptor-mediated nucleocytoplasmic transport. Thereby, plays indirectly a more general role in cargo receptor-mediated nucleocytoplasmic transport.

The mouse version of this antibody was raised by immunizing mice with human NTF2.
Application Notes
To characterize this antibody, a western blot on recombinant NTF2 protein was preformed using the mouse version of this antibody. Further, an ELISA on wild-type and C-terminal truncation mutants was preformed using this antibody. It only detected the wild-type protein not the C- terminal truncated proteins. Finally, the distribution of ran was determined in living BHK21 cells in the presence of this antibody. Normally ran is nuclear but with this antibody ran is redistributed to the cytoplasm (Steggerda et al, 2000; pmid:10679025).

Note on publication
It is shown that cytoplasmic injection of anti-NTF2 mAbs resulted in a dramatic relocalization of Ran to the cytoplasm. This provides the first evidence that NTF2 regulates the distribution of Ran in vivo.
