The original antibody was generated by immunizing chickens with phosphorylated tau peptides conjugated to KLH, constructing an scFv phagemid library from spleen and bone marrow RNA, and selecting specific binders through ELISA screening of the phage library against biotinylated phospho- peptides.
Application Notes
The specificity of the original antibody (chicken scFv, lambda) was evaluated via an ELISA. A chimeric human IgG1 version of the antibody was generated. The chimeric variant preserved the original binding characteristics as demonstrated in an ELISA. The binding affinity (KD) of the chimeric antibody was measured at 2.045 µM via SPR (Shih et al., 2012; PMID: 23148212).
