Specificity
This antibody is specific for the Streptococcus pneumoniae Pep27. The epitope recognized by this antibody is located within the residues 18-27, with key residues crucial for the binding being Y24, R26, and K27, as identified through alanine scanning and further supported by molecular docking analyses. Streptococcus pneumoniae Pep27 is a small, secreted peptide consisting of 27 amino acids initially described as a "death signal peptide".

Immunogen
The original antibody was selected from a synthetic human scFv library encoding HA-tagged scFv clones through five rounds of biopanning against synthetic and recombinant Pep27.
Application Notes
The dissociation constant (Kd) of the original human scFv version of this antibody was 1.1 µM as measured in an ELISA. This antibody specifically detected Pep27 in an immunoblot and in an environment mimicking in vivo conditions, demonstrated in human serum (Kim et al. 2018; PMID: 29753735).
