Specificity
This antibody binds a conformational epitope on the human PSMA. This antibody can bind native PSMA protein but were unreactive with any fusion protein containing a portion of the PSMA sequence. Heat denaturation of PSMA also results in loss of binding.

Immunogen
The original antibody was generated by immunizing mice with cell membranes and purified PSMA protein isolated from LNCaP cells.
Application Notes
This antibody was capable of detecting membrane fractions from human prostatic adenocarcinoma LNCaP and PC3 cells in a solid phase ELISA. This antibody was used for immunoprecipitation of PSMA from LNCaP cell detergent lysates. Antibody binding competition assays were done using flow cytometry and it revealed that 7F12 binds an epitope closed to another murine anti-PSMA antibody 3C6. This antibody was shown to mediated antibody dependent cellular cytotoxicity in human prostatic adenocarcinoma LNCaP cell targets with PBMC's from two donors (WO2003064606). This antibody was also used in the treatment of in vivo tumor xenografts with a combination of Taxotere® (US20090297438).
