This antibody is an affinity matured version of the murine parent antibody 3C4F5. This antibody was generated by panning against a antibody phage library created using PCR mutagenesis.
Application Notes
The binding characterization of the antibody was done using ELISA. To show the effectiveness of this antibody, a competitive immunoassay was performed on the serum of unknown patients using the human version of this antibody. It was compared to a commercial testosterone kit. The results were comparable (WO1999012974). The original Fab fragment of this antibody, having 5 optimised CDRs, showed 32-fold higher relative affinity to testosterone, 60-fold lower cross-reactivity to DHEAS, 2 fold lower cross-reactivity to 5α-DHT and over 9-fold lower cross-reactivity to androstenedione compared to the original monoclonal antibody (Ari Hemminki, 1998; ISBN 951-38-5343-8).
