Immunogen
The original antibody was selected from a llama-derived nanobody phage display library by screening against biotinylated TNF-alpha.
Application Notes
The original antibody (llama VHH) was screened for affinity to its target via ELISA and identified as antagonistic through functional assays, such as MTT cytotoxicity assays. A bi-valent version of the antibody (termed VHH#3F) was used in an in vivo model for IBD. Treated mice exhibited increased colon length and improved histological scores compared to the negative control (PBS-treated mice), indicating a reduction in inflammation and tissue damage as well as an overall therapeutic effect on chronic colitis symptoms (US9371381B2).
