Specificity
This antibody binds tritrpticin (VRRFPWWWPFLRR), which is a member of the cathelicidin family, a 13mer tryptophan-rich group of diverse cationic antimicrobial peptides found in neutrophil granules. It has a broad spectrum activity against a range of microorganisms, fungi, and protozoa.

Immunogen
The original antibody was generated by immunizing female BALB/c mice of 6-8 weeks of age with 200 µg of K-tritrpticin-diphtheria toxoid conjugate emulsified with equal volume of complete Freund’s adjuvant.
Application Notes
The binding characterization of this antibody was done using ELISA. In an indirect- binding ELISA where the peptides were directly immobilized on to the ELISA plate, mAb 6C6D7 could bind to tritrpticin and its N-terminal deleted antibacterial analogs at nanomolar concentrations. The binding affinity of this antibody to K-tritrpticin was analyzed using surface plasmon resonance and it was reported to be Kd= 350 nM. The binding of 6C6D7 to tritrpticin is characterized by negligible change in ΔGeq with respect to temperature that suggests that the antibody does not undergo any major conformational alteration during the binding of peptide antigen. This antibody was also used to bind tritripticin and related peptides to further analyze their anti-microbial activity (PMID: 24086578).
