Application Notes | The specificity of the antibody for IR was determined by ELISA analysis. The binding affinity of the antibody to IR-A and IR-B was measured by surface plasmon resonance (Kd= 2.57 and 2.98 nM respectively). The antibody blocked insulin induced IR phosphorylation in a dose-dependent manner on HuH7 cells. The antibody effectively decreased insulin induced glucose uptake at concentrations of 1 nM and 10 nM in vitro. The antibody induced hyperglycemia in C57 mice in in vivo experiments. |
