The original antibody was generated by panning a non-immune human scFv-phage display library against CA IX-expressing cells (293T-G250 cells).
Application Notes
This antibody is an Ab04787 [G10] variant designed with specific mutations (H310A/H435Q) to improve pharmacokinetic properties and serum persistence. The specificity of this antibody's original format (human scFv) was validated via ELISA, demonstrating its selective binding to CA IX-expressing cells. It was further confirmed through flow cytometry and cross-competition studies. This antibody exhibited an IC50 of 13.53 µg/ml to stable G250-expressing 293T cells. This antibody was tested in vivo in Ncr Nude mice (athymic mice) bearing luciferase-expressing tumors to assess its efficacy in targeting CA IX, exhibiting mild (~20%) inhibition (US8466263B2).
