Specificity
This antibody binds the extracellular domain of the human CD47 and cross reacts with cynomolgus monkey CD47. CD47 is broadly distributed on normal adult tissues, as well as ovarian tumors, and is especially abundant in some epithelia and the brain. IAP or integrin associated protein has a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It further plays an important role in memory formation and synaptic plasticity in the hippocampus. It also acts as a receptor for SIRPA. Binding to SIRPA prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells.

Immunogen
The original antibody was generated by immunizing a Bactrian camel with hCD47(ECD)-Fc fusion protein and the peripheral blood cells were used to generated a library. The particular clone was identified by screening the library for blocking the interaction between hCD47(ECD)-Fc and hSIRPa(ECD)-Fc-Biotin.
Application Notes
This antibody can effectively block the integration between human CD47 and SIRPa. The binding characterization of this antibody for CD47 was done using flow cytometry. The binding affinity of the VHH-Fc antibody for hCD47(ECD)-Fc was determined to be Kd= 4.92 × 10^(-10) M (WO2018233575).
