Specificity
This antibody is specific for human C3b. C3 plays a central role in the activation of the complement system. Its processing by C3 convertase is the central reaction in both classical and alternative complement pathways. After activation C3b can bind covalently, via its reactive thioester, to cell surface carbohydrates or immune aggregates.

Immunogen
The mouse version of this antibody was raised by immunizing a mouse with human C3b and iC3b bound to a carbohydrate matrix via the alternative pathway of complement activation.
Application Notes
To show that a monoclonal antibody directed against the merozoite and human polyclonal IgG from merozoite vaccine recipients enhanced RBC invasion in a complement-dependent manner, immunofluorescence was performed on merozoites with RBC using the mouse version of this antibody (Biryukov et al, 2016; pmid:27333049). Flow cytometry was performed on intact and lysed PNH erythrocytes using the mouse version of this antibody to analyze the deposition of C3 (Lindorfer et al, 2010; pmid:20068220). While studying the role of the complement system in childhood malaria, immunoprecipitation was performed on erythrocytes using the mouse version of this antibody. Furthermore, a western blot was performed on erythrocytes using this antibody (Pawluczkowycz et al, 2007; pmid:17911641). To examine the ability of several commercially available phycoerythrin (PE)-labeled anti- Mouse IgG antibodies to bind to B cells opsonized in milieu which allow or preclude complement activation, flow cytometry was performed on Raji cells, Z138C cells or CLL patient B cells using the mouse version of this antibody. Furthermore, Raji cells, Z138C cells or CLL patient B cells were stained for immunofluorescence using this antibody (Beum et al, 2004; pmid:15604014).

Note on publication
The ability of several commercially available phycoerythrin (PE)-labeled anti-Mouse IgG antibodies to bind to B cells opsonized in milieu which allow or preclude complement activation was examined. Even when large amounts of fluorescently labeled RTX are bound to the cells, binding of the anti- Mouse IgG probes is substantially inhibited if C3b is deposited on the cells.
