Specificity
This antibody is specific for the pep27 protein of Streptococcus pneumoniae. Specifically the epitope Y24, R26 and K2. Pep27 is a secreted peptide which is the signal sensed by VncR/S.

To generate this antibody a synthetic human scFv library encoding HA-tagged scFv clones was used for biopanning against pep27.
Application Notes
An western blot was performed on proteins from S.aureus using the scFv version of this antibody, it showed that the antibody was specific for the pep27 protein. Furthermore an ELISA was performed on His6-MBP-Pep27, it showed specificity of this antibody to pep27 (Kim et al, 2018; pmid: 29753735).

Two single-chain antibody variable fragments (scFv) were screened using a phage display from a large human synthetic scFv library to select clones E2 and F9. Dissociation constants (Kd ) of E2 and F9 were 1.1 mM and 0.50 mM, respectively. E2 and F9 did not cross-react with other pneumococcal and unrelated proteins.
